Typical purity is >90% CD3+ T cells as assessed by flow cytometry. Sub-populations (CD4+, CD8+) are further enriched by immunomagnetic positive selection, with purity typically >85-95% depending on the specific marker and species. A Certificate of Analysis with purity data is included with each lot.
Many anti-human CD antibodies cross-react with cynomolgus and rhesus monkey T cell markers, including CD3, CD4, CD8, and CD28. Cross-reactivity varies by clone and species. We can provide guidance on validated antibody clones for your species of interest.
Thaw rapidly in a 37°C water bath until a small ice crystal remains. Transfer dropwise into pre-warmed complete culture medium. Centrifuge at 300g for 10 minutes, aspirate supernatant, and resuspend in fresh medium. Allow 2-4 hours of rest in culture before functional assays for optimal viability and activation response.
Post-thaw viability is typically ≥70% for cryopreserved T cells. Viability is assessed by trypan blue exclusion or flow cytometry (7-AAD/Annexin V). Fresh cells are shipped with viability ≥90%. Recovery and function can be improved with overnight resting before stimulation.
