Vitreous humor is collected via pars plana vitreous tap using a 23-25G needle under general anesthesia, or during vitrectomy at necropsy. The procedure is performed by trained veterinary staff or ophthalmic surgeons. In terminal studies, both eyes are typically sampled.
Typical yield is 0.5-2.0 mL per eye depending on species size. Cynomolgus and rhesus monkeys yield approximately 1.5-2.0 mL, while marmosets yield approximately 0.3-0.5 mL. Pooled samples from multiple donors are available for larger volume needs.
Vitreous humor is evaluated for clarity and absence of visible hemorrhage. The gel-like consistency is maintained during collection. Samples are centrifuged to remove cellular debris before aliquoting. Samples with significant blood contamination are excluded from standard inventory.
