Buccal swabs are collected under light sedation using a cytology brush or flocked swab. The swab is firmly rubbed against the inner cheek mucosa for 30-60 seconds to collect epithelial cells. Food is withheld for at least 30 minutes prior to collection to reduce contamination.
A single buccal swab typically yields 0.5-5 µg of genomic DNA, sufficient for multiple genotyping assays or SNP arrays. Two swabs are recommended for whole-genome applications. DNA quality is suitable for PCR, microarray, and next-generation sequencing.
Swabs are placed in DNA stabilization buffer (room temperature stable for days to weeks) or dry-shipped. Extracted DNA is stored at -20°C or -80°C. Stabilized swabs can be shipped at ambient temperature; extracted DNA ships on dry ice.
