Cytosol contains soluble Phase II drug-metabolizing enzymes including glutathione S-transferases (GSTs), sulfotransferases (SULTs), N-acetyltransferases (NATs), aldehyde dehydrogenases (ALDHs), and alcohol dehydrogenases (ADHs). It does not contain membrane-bound CYP enzymes (those are in microsomes).
Store at -80°C. Avoid repeated freeze-thaw cycles. Thaw on ice immediately before use. Protein concentration is typically 20-50 mg/mL. Dilute to working concentration in appropriate buffer for your enzyme assay.
Cofactor requirements depend on the target enzyme: GSH for GSTs, PAPS for SULTs, Acetyl-CoA for NATs, NAD+ for ALDHs. A cocktail of all cofactors can be used for comprehensive Phase II profiling.
