Liver microsomes contain the full complement of membrane-bound drug-metabolizing enzymes including CYP1A2, CYP2C9, CYP2C19, CYP2D6, CYP3A4/5 equivalents, UGTs, and FMOs. CYP activity characterization data for major isoforms is available on request. Activity levels vary by species and individual donor.

Store at -80°C. Avoid repeated freeze-thaw cycles; aliquot upon first thaw if multiple experiments are planned. Thaw on ice immediately before use. Dilute to working concentration in incubation buffer just prior to the experiment. Microsomal protein is stable for years at -80°C when stored properly.

Standard protocol: 0.5 mg/mL microsomal protein, 1 mM NADPH (or NADPH-regenerating system), 100 mM potassium phosphate buffer (pH 7.4), 37°C incubation. Typical time points: 0, 5, 15, 30, 60 minutes. Adjust protein concentration based on CYP content for species-specific optimization.

Both individual donor and pooled donor (typically 3-10 donors) microsomes are available. Pooled preparations reduce inter-individual variability. Individual donor microsomes are useful for CYP phenotyping and genotype-phenotype correlation studies.

Each lot includes: total protein concentration (BCA/Bradford), CYP content (CO-difference spectrum when available), and a Certificate of Analysis. CYP isoform-specific activity data (using probe substrates) is available on request for an additional characterization fee.
