S9 fraction (post-mitochondrial supernatant) contains both microsomes (Phase I enzymes: CYPs, FMOs) AND cytosol (Phase II enzymes: GSTs, SULTs, NATs). It provides a more complete metabolic profile than microsomes alone. S9 requires cofactor supplementation (NADPH + UDPGA + PAPS + GSH) for full Phase I + Phase II activity.

Store at -80°C. Avoid repeated freeze-thaw cycles; aliquot upon first thaw. Thaw on ice immediately before use. S9 protein concentration is typically 10-30 mg/mL. Dilute to working concentration (typically 1-2 mg/mL) in incubation buffer.

Standard Ames test S9 mix: 10% v/v S9 fraction in cofactor-supplemented phosphate buffer (NADPH, G6P, G6PDH, MgCl2, KCl). S9 activation mimics mammalian hepatic metabolism for the bacterial mutagenicity assay. Follow OECD 471 guidelines for specific concentrations.

Both individual and pooled donor S9 fractions are available. Pooled preparations (3-10 donors) are standard for regulatory studies to represent population-average metabolism.

Each lot includes: total protein concentration, and a Certificate of Analysis. Enzyme activity data for major CYP isoforms is available on request.
